Morphological Analysis Of Organoids

Seattle Genova offers alteration of organoids morphology by manipulating the condition of its cultures. We believe that the morphology of an organoid often resembles the original organ and provides a mechanism that governs a tissue-specific constraint on organoids. We provide various frameworks to identify these constraints on the organoid morphological features by quantifying them from a wide range of perturbations by microscopy images.

Colorectal Organoid Morphology Analysis 

Seattle Genova offers organoids that allow precise ex vivo testing of various tumors. Our system focuses on the systematic dissection with compound effects on the 3D organoids, by developing high throughput imaging and other quantitative approaches. The general morphology of our patient-derived organoids (particularly from Colorectal cancer patients) was treated with >500 small molecules and captured 3 million images by confocal microscopy. Our software network like SCOPE is utilized to measure compounds induced in the reorganization of organoids. Seattle Genova also has expertise in the analysis of specific phenotypes which is induced by compound classes and found various inhibitors to dissemble PDO through focal adhesion bloating by enhancing stemness. Through viability classification, the heterogenous susceptibilities of PDO to various clinical drugs.  

Kidney Organoid Morphology Analysis 

Seattle Genova utilizes Madin-Darby canine kidney cysts organoids which obey several constraints on certain parameters. The number of cells increases with increasing the size of cysts and can be altered by varying the condition of the culture. Our quantitative framework is utilized for identifying constraints on organoid morphologies which may inform future efforts to engineer various organoids. 

Rectal Organoid Morphology Analysis (ROMA)

Seattle Genova developed various physiological cystic fibrosis transmembrane conductance regulators (CFTR assay) which are based on the morphological difference between the rectal organoids. ROMA can be utilized as a standard to obtain the size of organoids. 

Cerebral Organoid Morphology Analysis

For cerebral organoids, Seattle Genova utilizes a high-resolution imaging pipeline to image cytoarchitecture information and spatial localization of cells. The localization and quantitative analysis of cortical layer markers also reveal the heterogeneity of cerebral organoids.

Heart Organoid Morphology Analysis

Seattle Genova also offers a customized spectral-domain optical coherence tomography system to characterize the growth of human heart organoids. The development of chamber structures and corresponding patterns could analyze through optical coherence tomography, to demonstrate the 3D images in a fast, non-destructive, and label-free manner.
