Production of Peroxisomes Proteins

In most eukaryotes, peroxisomes are membrane-bound structures that play an important role in lipid metabolism as well as the transformation of reactive oxygen species like hydrogen peroxide into harmless molecules like oxygen and water.

The phospholipids bilayer that makes up these structures contains numerous membrane-bound proteins, which include those that function as protein transporters and translocators. However, there is also some reason to link peroxisome-resident enzymes to ER-mediated protein biosynthesis.

In general, one of two signal sequences is found in enzymes and proteins that are targeted for the peroxisome. That is, there are brief intervals of a few amino acid residues that identify the protein's sub cellular position. The Peroxisome Targeting Sequence 1 is the most prevalent signal pattern (PTS1)

A 9-amino acid N-terminal signal sequence may also be included in peroxisomal proteins. This sequence contains two dimers that are spaced apart by five amino acids. Leucine and arginine combine to form the first dimer, while histidine and leucine combine to form the second dimer. The single- letter amino acid coding for this signal sequence is RLx5HL.The phospholipids of the peroxisome are mostly synthesized in the smooth ER. A peroxisome can split into two organelles as it expands in size as a result of the entry of proteins and lipids.
